The Human Blood RNA Purification Kit provides a rapid, simple and effective approach to isolate the total RNA from human whole blood. The process is based on a spin column format. The procedure involves cells disruption and homogenization in the presence of guanidinium thiocyanate. After adding ethanol, RNA binds selectively to a silica membrane. Residual contaminating genomic DNA is digested by DNase I, which is applied directly in the column. After a series of rapid “wash-and-spin” steps to remove contaminating cellular components, the RNA remains bound to the membrane. Finally, nuclease-free water is used to elute the RNA from the membrane. Note that RNA shorter than 200 nt, such as 5S RNA, tRNA and microRNA, do not recover efficiently with this system.
